Kinetic Analysis
The Octet® R2 System monitors binding events in real- time measuring binding association (ka) and dissociation (kd) kinetics, and affinity constants (KD). The system’s two channels can be used independently for screening purposes, or in tandem, pairing the sample read with a dedicated reference for high-quality kinetic
characterization.
Samples are loaded in standard microplates, eliminating fluidic line maintenance and expanding the number of samples that can be run in an assay. Pre-defined templates in Octet® BLI Discovery software streamline setup prior to running an assay and minimizes training needs. The software also offers users a range of parameters and metrics for analyzing acquired data in a powerful yet intuitive software interface.
Quantitative Measurements
The 2-channel Octet® R2 system directly detects the presence of specific proteins in solution with minimal interference from complex matrices. Accurate and reproducible concentrations can be determined in as little as 2 minutes per sample using a simple, one-step assay.
Sample plate cooling for temperature-sensitive proteins
The sample plate temperature can be controlled from 15–40°C, which enables reliable kinetic determinations at different temperatures for temperature-sensitive proteins. Additional advantages of sample cooling include the ability to rapidly determine binding rate constants at multiple temperatures to extrapolate thermodynamic parameters.
Biosensor Re-racking
The Octet® R2 system provides the ability to re-rack biosensors at the end of an experiment. Biosensor re-racking provides enhanced flexibility in loading ligands on biosensors and reduces operational costs when biosensors are loaded with precious target reagents.
